I have an Asus P5K motherboard and have three extra PCI slots to spare. The PCIe express port is lready taken with my graphics card and I am thinking of adding another to my system. I am a gamer as well as a Graphic designer, and the main manufacturer of my graphics processors is Nvidia (I currently have an Nvidia GeForce 8800 GS with 512mb of video RAM). I am thinking of adding a Zotac NVIDIA GeForce GT 610 512MB GDDR3 PCI Video Card in one of the three extra PCI slots I have on hand. Would this be possible to have a GT 610 and a 8800 GS running on the same system?

Yes they all run the same driver so it would work. It would allow you to run the main screen for gaming at full power and the (older card, or in your case smaller card) could run extra monitors for email, or Skype, or team speak, or whatever.

I would not try to use the older/smaller card for physix. It will actually slow the system down.

The 5000-6000-7000 NVidia cards are to old to work well. I recently tried to add a 5000 to my gtx470 rig just to run an extra monitor for coretemp, afterburner, and steam friends list while in surround. Windows got very mad at me and I had to remove the card and reinstall the newest drivers.

The extra card may not give you more power as such. The 610 is a rather slow card. Much more entry level. To run extra screens that is just fine, but I do not think it will help your games.

If you want to game, Most newer cards will let you game and many can run 3 or 4 screens(I would aim GTX 650 ti[if the price is right for you] for med/high settings at higher screen resolutions and more to the 660/660ti or faster for a better gaming experience.).

This all assumes your cpu is not too old and holding back newer cards performance.

Again, if you are happy with the current performance and just want more screens, the 610 should be fine. Also, some games should be run in Windowed or Windowed fullscreen(fake full screen) if they interfere with what is on the second screen while playing.

If you are looking for more graphics performance, then you need to get a higher end graphics card. Both of the cards you listed are low end by today's standards. I would suggest you take out the older GeForce 8800 GS 512MB card and replace it with something newer like a GTX 650 or GTX 650ti. The newer card should be able to run 2 monitors on its own without the hassle of trying to use two graphics cards. Take a look at the Graphics Card Hierarchy Chart for more information.
