No, PCIe 3.0 is backwards compatible with PCIe 2.0. The only time that PCIe 3.0 would provide a significant benefit would be when running three or four graphics cards in SLI or Crossfire. Even then the difference isn't more than a few frames per second.

There are gains when using a PCI-E 3.0 slot over a PCI-E 2.0 slot.

As they conclude in the comparison it does not make a big enough difference to really notice though.
