Yes.

I'd guess that you position the fan opposite the CPU cooler fan, and that the CPU cooler is a traditional (stock or similar) cooler? Because that way, you can have the side fan assist the CPU fan in transporting the hot air from the CPU out of the case (side fan set to exhaust), or you can set it up so that it works hard to keep the hot air IN the case (side fan set to intake). The latter would probably make a lot of noise.

Fan is right across GPU so I placed it as exhaust. I hope that is right. :) (its pushing hot air out now)

Depends on the cooler. Reference based coolers for the GPU it won't be good, infact it would be bad for airflow, as you're taking away the air it requires, since it pushes the air out of the chassis. You would only want to do that with non reference coolers, and even then, I'd be careful that you don't overheat yourself with all the heat coming out

GPUs have their air intake opposite from the CPU, so unless you operate in a near vaccum, or the air intake fans/filters at the front of the cabinet are blocked, there's no chance of 'starving' the GPU cooler by forcing hot air out elsewhere in the cabinet.
