is PCI express x16 MOBO interface compatible with PCI Express 2.0 x4 RAID Controller card?

The question should be addressed to the RAID controller manufacturer. You want to know if it's compatible with your motherboard and if it supports drives larger than 2TB. It should be compatible with the PCI Express slot, but it probably is not supported by the motherboard.
