I just bought a new PSU today, and after I plugged it all in, I heard a faint crackle and smoke puffed out of my Optical drive, I don't know exactly what happened, and I'm worried that I may have also fried my HDD, since they are on the same cord.

Does anyone know what might have happened?

I tried booting with the old PSU and it did not smoke, but it booted into the "ASUS EZ mode UEFI Bios"

Does this mean that the HDD is definitely blown? Is there some way to check using the UEFI interface?

If the drive is still working it should be picked up and reported in the bios.
However it does not mean it is fully working if the case.
A boot into windows and an insert of a cd or dvd to see if the drive reads the disk would be your next step.
If none of these work, I think you will know the answer.
Obviously there was a short or one created when you plugged the new power connector into it from the new PSU.
You have to ask how cheap was the new PSU this may of been the cause.

Well I feel like a moron now, I found the problem, and its not the PSU, I somehow managed to connect the cords for the HDD and Optical Drive into the PSU's PCI-E Pin slot, thus overloading the drives.

On the plus side you can now use your optical drive to cook pop tarts. Just looking at the brighter side. Well at least a optical drive and a hard drive are not too expensive relatively.

Why use a toaster when you can use ruined computer parts?

It's not too big of a problem, since this was just a gaming pc, I use my laptop for all of my school stuff. It's just gonna be a pain to reinstall all of my steam games and other stuff.
