I have 2 Cat5e wall jacks, main floor and basement. My FIOS router (Actiontec Rev I) is on the main floor. I have my TV and Blu-ray player hardwired to that router. I plan to run a cable from the WAN port to the wall, couple the other ends of the wall jacks in the smart box, go out of the basement wall jack, into the basement router (TP-Link TL-R860). The basement router would feed a computer, xbox, blu-ray, etc.

Would all I need to couple the connection is the below?

You don't need another router. Run a cable between the FIOS router' LAN port, through the wall between the floors, into simple 5/8 port 1Gb switch.

As for these couplers - it depends. I have seen some of them made poorly, where even 10Base-T had a problem running. You will be better just getting longer cable, or asking someone to make one for you.

I agree about the switch rather than a router. You should be able to place the coupler in the box as you describe. Many times people place a switch here also to run all the cables in the house. A simple coupler will be simpler though if you only have 2 wall jack you want to activiate. Be careful that all 8 wires run though and they are on the correct pairs. Sometimes this is telephone wiring not data but you likely can make it work
