As you may have figured out by the title, I was wondering whether a 3.5" hard Drive would work in a 5.25" enclosure such as the Vantec Nexstar DX.

In theory it should work, provided that I add some 3.5" to 5.25" brackets to the hard drive.

Is there any reason why this won't work? The reason I ask, is I can get a 5.25" SATA USB enclosure for less than half the price of a 3.5" SATA USB enclosure.

About you get one 2.5"/3.5"/5.25" SATA/IDE to USB 2.0 Adapter (the UBS3.0 version). Then you can use it for all format HDD.
