I have a new laptop with 16GB Ram and at random my system spikes to 100% of system memory in use.
Not cached, In use and says 507MB is available

Process's in - sorted to most memory: http://imgur.com/tKTbBCd,VgcQJQY#1

My laptop is a HP Envy 1205dx
A10 - 4600M
ATI 7660G 3GB
16GB Corsair 1600Mhz (2x8GB)
Windows 8.1 64Bit

In the process when this happens my laptop gets super laggy and i can hardly move my mouse.
So i restarted my laptop and went back to task manager and it was still at 15GB in use.
I turned off the laptop and turned it back on an hour later it still showed at 15GB in use and then my system turned off and the screen went black.

When you turn off the laptop it resets the memory because it stores data for quick access on the memory and it didn't reset itself.

This happens at random but it does this quite often now and its very annoying.

Nothing 'stayed' in memory while your system was powered down.
When you resumed operation, all of the programs resumed what they were doing before they were interrupted and that is why it looks the same.

Make sure you "reboot" from time to time. Don't just put your computer to 'sleep' or 'hibernate' or suspend. That will clean things out.

From your picture, I can see that Firefox has a lot going on. It could be updating itself, or you could just have too many tabs open at the same time. Shut Firefox down and restart it.
