How to get an HDD ready for my new build. Do I need to format it and assign it a letter?

Gotta be honest here I've been messing with 2 different harddrives here and I dont know what I'm doing. I did all sort of things to 1 of the HDD's I but long story short I got it to work lol. Anyway I have a 1TB HDD here that I'm gonna start messing with. It was healthy but I went ahead and deleted partition anyway cause it wouldn't let me assign it a letter. Well now all I can do is create a partition. If I want to use this HDD on my new build next week do I need to do this (create partition, format, etc) or do I leave it alone? Windows is going to get installed on this HDD.

can I do all that in the BIOS?

I'm not sure what you mean by custom version of windows install. I assume you mean how when you install something it asks you if you want the recommended install or a custom install.
