Not enough standoffs for replacing motherboard.

My current motherboard that i have at the moment (Asrock N68-VS3) has Six standoffs and the motherboard that i have just ordered, along with a new processor, Graphics Card and Power Supply, requires 8 Standoffs.

The motherboard that i have just ordered is a MSI Z77MA-G45 and I was wondering whether i would be able to get away with using 6 standoffs?

Also when replacing a motherboard do you have to delete the existing OS?

You might get lucky and be ok using few standoffs but I wouldn't recommend it. You run the risk of causing the board to flex when your inserting or removing your components which could damage the delicate traces. I might try that as a last resort but I would urge you to do whatever you have to do to find some additional standoffs.
Personally, I usually do a slash and burn reinstall of my OS when I install a new motherboard. You could probably get away with not doing it but it gives me piece of mind to know that I will have the full functionality of my new board, and that I don't have too many pieces of code from my old one still living on my hard drive.
