PCI Express Sata-600 card expansion on a motherboard that natively only has Sata-300 slots, would it work?

I'm buying a new harddrive that has a Sata-600 connection. I know that they are backwards compatible and I accept the fact that the speeds would be reduced however I went looking for a solution.

If I add a PCI Express card to my motherboard that creates two Sata-600 slots would it work with the optimal speed of the hard drive?

If it is a HDD and not a SSD then it will not benefit enough from a SATA 6Gbps connection. The best HDDs only push about 250MB/s, which is within the SATA 3Gbps specification. Only a SSD would really need a SATA 6Gbps connection.
