Highest RAM on slot1 ?

I have 3gb ram (1gb and 2gb) ddr2, in my case should I put 1gb or 2gb on slot 1 ? I'm not sure if that will make a difference in speed

No, if the memory sticks match in speed. then it will not matter what slot you put it in.
I think your asking if you put a 2gb stick in with a 1gb stick in the second slot.

It does not improve speed just memory capacity.
Computers like more memory.
Because it allows you to load more things into the memory space.

When you run low on memory space things slow down because windows uses what we call a swap file.

A swap file is a part of the hard drive it uses to temporary store data if it needs to be retained.
If your system memory is low, and you wish to run another program.

Swap files are 100% more slower in speed than a ram memory module.
So to avoid swap filing in windows you add more system memory.
