I recently purchased a ASRock B85M Pro4 Micro ATX LGA1150 Motherboard and G.Skill Ripjaws X Series 8GB (2 x 4GB) DDR3-1600 Memory. The installation guide for the motherboard has a priority of 1 through 3 on the memory configuration table with priority 1 having slot a2 and slot b2 populated. So am I right in thinking those are the two slots I should put my two sticks of ram in for the best performance?

If the manual says a2 and b2 then that's where you should put it. Generally, it doesn't matter if you do a1/b1 or a2/b2 first as long as it's one of those. If you were to do a1/b2 then it would be running in single channel mode instead of dual. Just follow the manual directions and you'll be fine.
