MSI Radeon R9 270 / ASRock B85 Pro4 Motherboard Conflict?

Built my first computer over the weekend, used pcpartspicker to verify compatibility:

I assembled everything outside the case, computer did not post, received 5 beeps (process error). I removed the video card and it booted right away to bios screen. I continued to mess with the card and I did eventually get it to boot to bios with the video card installed.

I installed everything in the (including the video card), computer did not post, again 5 beeps. Removed video card, computer posted and proceeded to install Win7 and update it along with all updated drivers for motherboard.

Reinstalled video card, didn't post, 5 beeps. Dinked around with it (removing and reinstalling the card several times), computer eventually booted up like nothing was wrong. Installed updated drivers and software for the video card.

Moved the computer to my office, computer didn't post, 5 beeps. Turned off and turned back on, computer booted (didn't touch anything inside the case).

What could be the problem here? I am positive the card is seated in the PCIe slot, because I assembled the components outside the case and it only sometimes worked then as well. I wrote MSI customer service and received a canned response of making sure the power supply is at least 550W, card is seated, etc. I did not buy a 500W PSU because to me it didn't look like I needed one. Once the computer does boot I don't have any errors, and Windows runs fine. Installed Starcraft 2 and it runs smooth as butter on Ultra. Any help or suggestions are appreciated, thanks for your time!

Is there another pci e slot you can install the card in to find out if the slot is defective.

I put it in the second PCIe slot and when I turned it on it did nothing, no post, no beeps, nothing... just sat there with the fans running.
