My brother is hoping to upgrade his pc, but he or I do not know what would be the best parts to upgrade. His specs are
cpu-intel core i3 530

gpu- Gtx 260

mobo- asus P7P55D-E

Ram- 2gb of dual channel DDR3 ram

psu- antec 750W

This pc was built 8 years ago so it is outdated but we were hoping we would not have to start from scrap. If we upgraded the gpu to an amd r7 265 would mantle help boost the weak cpu? or would that gpu bottleneck the cpu too much? Can this mobo even support the r7 265? all help appreciated. The biggest question I want answered is, if he were to get the r7 265, would he be able to run multiple programs faster and better?

This PC is not that bad out of date, i3 processors were not even made before January 2010, so at best it is a little over 4 years old.

To get the most out of this build, you need to be running a 64 bit OS, 4 gig of RAM is a bare minimum, 8 gig is the sweet spot.
The GTX260 however is from late 2008, and indeed is behind the curve if you play games. A new graphic card would be a great addition for gaming, however it won't do any more than your current GTX 260 is doing now for normal programs and apps. That is where more RAM will help, and of course a faster processor if want to shell out the money.

For gaming, upgrading the gpu would help quite a bit. What OS are you running? 32 bit or 64bit? If 32 bit, you would be limited to only 4gb of memory, where as, if you are running a 64bit OS, you could go up to the max for your mb, which is 16gb. Going from 2 to 4gb ram, will help speed things up, nonetheless.

Here is a cpu that fits the socket of your mb,

not sure if you want to upgrade or save for a whole new system
