My motherboard has a single usb 3.0 port from the early days of 3.0 (early 2010) and it also has esata II. Which is better for a 3tb external drive used for backups? 3.0 drives are cheaper and more available, but I could get a hdd+esata/usb enclosure for not much more.

ESATA is always the preferred option, however USB 3.0 is faster than a HDD anyway so it really doesn't matter. I'd still pay a few bucks more to have ESATA/USB 3.0. its more flexible
