How to tell if the ram slot works properly

How can I tell if my ram slot is damaged Or not working properly?

If I use memtest86 on a ram on each of the slots and it passed, does this mean that the ram slots are working properly?

That would satisfy me, yes. Do you physically damage one of your DIMM slots?

No, I'm just making sure that my ram slot is working properly. So this just means that the new ram stick that I bought is faulty

If you use the same DIMM slot for each stick of RAM, and test it for at least one full pass, any errors at all represent a faulty stick. Technically, you'd want to use the DIMM slot that is designated as the 1st slot; the slot the MB manual says to use when just one slot is populated. (if in fact, your MB has that disclaimer)
