Is deleting temporary files in Windows 7 safe?

So I typed in %temp% in the start menu in the search field and saw a folder named temp. When I opened it and viewed the space the folder's contents were using it came out to be about 50GB. So I thought of deleting it but then thought of first consulting if it is safe or not. I use Photoshop, After Effects, Dreamweaver, Illustrator, Cinema 4D, Sony Vegas etc. as I am a graphics designer. So is it safe to delete these files? Will it affect the working of my Programs or OS etc.? Will it cause any problem? What cautions should I keep?

IMHO it should be safe to delete temp files , i have never had an issue doing so,

EDIT:i7 Baby raises a good point if you dont know what it is leave it , 50 gb is huge for temp files , sounds like you have a lot of game installs in there

well actually I don't have single game on my PC its just that I have a lot of PSD files and Tons of footage

For the files in the %TEMP% folder, so long as any applications are closed, there should not be a problem. Normally having some files in this folder should not present a problem, but I've found that various add-ins to Microsoft applications seem to cause this folder to fill up much more quickly, sometimes with gigabytes of files. While it's not necessary to empty it every day (though it doesn't hurt), it does help to keep an eye on the folder and empty it when it starts to build up its collection of garbage.

I've added a shortcut to this folder on my desktop, since its the one that affects the applications I use the most.

I find the best time to delete files in the %TEMP% folder is immediately after boot up. Normally, Windows and applications in general will not allow you to delete a file that is in use, so emptying it when no applications are open works best. And keeping that folder clean will have a direct impact on how fast some applications run, especially MS Outlook and MS Word.

Does it literally say ["xxx" was not found]?

Some application is causing those messages. Have you checked to see what applications are starting up when you boot your computer? You may have an application that writes its files to the %TEMP% folder. If you can find it, uninstalling and reinstalling it should resolve the problem. Just be aware of what files it writes to the %TEMP% folder so you do not inadvertently delete them again.

4 times? I think it's trying to get your attention! ;-)

It appears as though there's an ASUS utility trying to run...perhaps the System Update utility?
Perhaps a driver/app from the motherboard's utility disk? Some ASUS hardware?

Run MSCONFIG and click on the Startup tab to see what might be running from ASUS.
It may very well be something you don't need and you can turn it off from there.
