How Do I Backup A Constantly-Changing Large File To An External HD Periodically?

Hello! Normally, I am able to get all of my simple questions answered here or elsewhere on the web, but this question just proved too vague to get a quick answer.

All I am trying to do is regularly backup my Calibre Library (one file) on hard drive A to hard drive B. The Calibre Library changes regularly, so I'd like to back it up periodically.

The problem is that up until now, all I've ever done in terms of file management is copy my files en mass from one hard drive to another. I don't know how to update the copy of the Calibre Library (on hard drive B) without simply recopying the entire 800GB archive. I know there must be a simple way to do this, but I've never done it before, and as I said before: my question is vague enough to bury the requested answer in the back pages of Google.

In short: How do I UPDATE the backup copy of a large file (as the changes are insignificant by volume), rather than just laboriously RECOPYING the 800GB original to the other external HD?

Sorry, I've just never had to "tend" very large files in this way before. My Calibre Library is over 65K volumes. So I guess this is what "backing up" is!

get an backup software do a full back up, then set it up so every couple of days it will do a incremental backup. (An incremental backup is one that provides a backup of files that have changed or are new since the last backup)
