I think I found something: your new RAM is single side (SS), while in the official memory list there are only double side (DS) 4GB compatible DIMMs. Usually the SS DIMMs has memory chips only on one side, while the DS DIMMs has memory chips on both sides of the DIMM :

EDIT: it seems I was not right, because I...

Hello,

What RAM amount is listed in BIOS? Because, if you let Virtual Memory on Auto, a 6GB virtual memory corresponds to 4GB RAM installed, as in 2x2GB DIMM, and that means the 2GB DIMM is single sided and the 4GB DIMM is double sided. Please list the exact markings from the both DIMMs labels (maker/model).

For best compatibility you must use the memory brands/models listed in the Memory Compatibility List from the ASUS website:

http://www.asus.com/ph/Motherboards/P5G41TM_LX3/HelpDesk_QVL/

No Strontium memory is listed there and only a handful of 4GB DIMMs compatible (Kingston, Micron). If the 4GB is Strontium, try to return it and change it with Transcend, or do a full refund and buy a 2x4 GB Kingston kit. The best way is to go to the nearest PC repair shop and test some 4GB DIMMs.

Pc works fine with my 2Gb ram , i have been using it for few years
I have 2 GB strotium RAM and 4 GB of transcend RAM.

When you try to boot only with the 4GB one, the DIMM is inserted in the first memory slot? If so, that means it's incompatible with your motherboard. The 4 GB DIMM is new? If so, try to change to a Kingston DIMM (but I recommend you to go to a PC repair shop and test several DIMMs).

Yes the 4gb ram is new . So you mean to say that it wont work in my motherboard right?

As I was saying, if you tried to boot only with the 4 GB DIMM inserted in the first DIMM slot (the one closest to the CPU socket) and the PC is not starting, that means the memory is incompatible (or, less likely, DOA). Try to run the RAM at 1066MHz frequency.

Please list the exact specifications for both DIMMs (take some pictures for each DIMM label and post them here); in the manual, page 1-13, they say that DIMMs must be unbuffered and non-ECC.

EDIT: it seems I was not right, because I found out that your Transcend is apparently a DS DIMM... In the pictures it has 16 chips, so 16x256MB = 4GB, but it the motherboard's manual (page 1=13) it says it cannot support DIMMs with 256 MB chips or less, so for 4 GB you must have a DIMM with only 8 memory chips on it (8 x 512 MB). The best solution is to test several different DIMMs from different manufacturers, so a visit to a PC repair shop.
