I need to create a new PDF port on windows 7 but new port button DOESN'T open.

Under printer properties, you go to Ports and then to PDF PORTS and click on New Port, a new window need to be open to select a file, but the button do not open.

any one can help on this one?

True. The only other suggestion is the software vendor. See if they have a support forum. Problem here is your dealing with a very specialized application. Or try and Google the app name and PDF port. Someone out there must have something on it.

I have a software that create business quotes, after is done it need to create the quote as pdf and that is accomplish by the pdf port that i need to create so it can send the quote to that port and create as PDF and then send it by email with the pdf attached to the customer.

Try going to Add a printer in your Devices and Printers and select local. You may be able to create the new port from there even if you cancel the printer add. Any ports created are shared among all devices. I'm not 100% sure that will get you to where you need to be.

Is a PDF printer, it will ask me for the drivers and as you know pdf printers are virtual
