This is my motherboard: http://www.newegg.com/Product/Product.aspx?Item=N82E168...

I currently have 8 GB of this RAM: http://www.newegg.com/Product/Product.aspx?Item=N82E168...

I would like to add only 4 GB more, but is it better to add 2x 2 GB sticks, or just 1 4 GB?

EDIT: Also, upon searching Newegg, there does not appear to be an EXACT match in 4 GB...is this OK? I can find DDR3 1600 (PC3 12800), but for 4 GB, the CAS Latency is different (either 7 or 9, not 8 like my current sticks).

Always add ram in matched pairs if the board support it, unless you want to get to 32gb in the very short future

By matched pairs, are you saying that it is bad to have a pair for 4 GB sticks, and then to add a pair of 2 GB sticks?

No, that is fine. That's two different pairs. I like to have all the same but in all honesty my laptop has 4 different (similar) sticks and runs fine.

What I wouldn't do is add 1 2gb and 1,4 GB
Just check in the manual which sockets to use for each pair
