The motherboard must support SLI, or it simply wont work if you want to SLI 2 cards together.
Late Edit: I may have misunderstood what you are asking.... If you are not planning on running SLI, it does not matter if the board supports it or not.

Nothing is changing on your graphics card. Your non-SLI motherboard will only be able to accept one GPU. If you think you'll want to run SLI on your setup down the road then I'd advise finding a motherboard that supports SLI.
From the sound of it I don't think you understand what SLI is... SLI is when you have TWO Nvidia GPUs working together in your system. If you do not plan to get a second GPU you have no reason to get a motherboard that supports SLI.
