PCI-E is backwards compatible, it will work in either slot without any trouble at all. So to conclude it will work in both a PCI-E 2.0 slot and 3.0 slot, as well as even older generation slots.

GT 640 requires PCIe 3.0 slot it can be run on PCIe 2.0 slot althought it can be fit in PCIe 2.0 slot but what u will do if this doesn't work.

it doesn't "require" a 3.0 slot, it is just their recommended slot. With a card like that it makes no difference in a 2.0 slot at all, and to be true neither does it effect any other card. 3.0 slots only have a few extra added BIOS features, and no card is taken to a better advantage on 3.0 compared to 2.0 -
