Must a HDD be formatted when put in a new enclosure?

I have a 2TB drive that's been in an old computer. The disk is used for storage so It wasn't the primary disk. I have another external 2TB disk with the same data on it (around 1TB).
If I take the first HDD out of the computer and put it in my new USB 3.0 enclosure, does it need to be formatted before I can use it?
It would take a while to transfer all of those files from the other drive again.

Solution
Try it in a usb 2.0 port. Computer might not have 3.0 driver on it, or it could be a cheap circuit board adapter that does not do 3.0 correctly.

You can try a different drive, but im sure it has to do with the encolsure adapter and not the drive.

Thanks I inserted the HDD in my new enclosure and the USB cable in one of the USB 3.0 port on the back, but it isn't detected at all. It doesn't show up in Disk Management or Device Manager, I turned it off again and tried the other 3.0 port, nothing. Then I tried the front 3.0 port, but nothing happens.
Anything I can try else I can try?

Thank you again. Why did I not think of that..
It was detected in a USB 2.0 port and automatically installed by Windows. Then When I tried a 3.0 port again it worked there too.
I ran a benchmark in Samsung Magician and the Read speed is 104MB/s and the write 110MB/s, are those normal (max?) speeds for a 5400rpm HDD (WD20EARX) in USB 3.0?

For a 5400 rpm drive, those speeds are great.

5400rpm drives are slower then the desktops drives.

The drive itself is maxing at that speed, the fact that it is on USB 3.0 that can do over 600MB per second is irrelevent (putting 170mph rated tires on a minivan does not make it able to go 170mph).
