To increase GPU voltage must the motherboard be compatible? Will the tweaker not work otherwise?

Are you talking via MSI Afterburner or via BIOS changes?

AFAIK the GPU is independent of the rest of the motherboard - it's irrelevant what your motherboard is if you want to change the voltage of the GPU; but for the CPU it's a different story.
Same goes for changing clock speed for core and memory.
For fan speed also, compatibility doesn't matter. You can freely change your CPU and graphics card fanspeed irrespective of the motherboard; only requirement for the CPU fan is that it should be a 4 pin fan; not a 3 pin.
