Can the content of a second hard drive be merged with the first, leaving the second able to be removed?

I installed windows 7 64bit initially on a 40 gb hard drive, and 500 gb of mixed content on a 2TB wd Green drive.

I now have a 1TB WD Blue drive, I wish to put everything on, leaving me to use the WD green drive as an external drive.

So, my question is, can this be done? and if so would anyone be able to supply me with a step by step guide. I have NEVER backed up anything until this point other than installation files, so I am pretty new to backup/restore.

If it can be done within win 7 great, if not I am prepared to buy whatever software is needed (within reason)

ps. I mentioned backup/restore as I wish to be able to keep a copy of the backup on the WD Green (external drive)
