I want to change one of three antenna to a directional on a D-Link DIR 855 router

The first router you posted the answer would be yes because it only uses a signal antenna at a time.

The new routers that can do mimo (ie 300m or 450m) use all the antenna simultaneously. You CAN put a different antenna on these but you likely will break the mimo feature. It will unlikely be able to figure out how to combine the signals and you will be limited to 150m. Used to be my favorite trick to put bigger antenna on a router now you make any changes at all and you run the risk of actually degrading your speed.

Still doing what you want will work the router is smart enough to only use a single antenna without the mimo features without you having to configure anything.
