Can ASRock 980DE3-U3S3 handle a fx 6300 and gigabyte 4gb R9 270x

Yes, any motherbd with a pcie x16 slot can though. What you want to do is make sure your processor can feed the GPU (graphics card) properly and not hold it back. An fx-6300 can do that so you are set in that department too.

4gb GPU would be useful for multiple screens or high res single screen, otherwise go for the 2gb model.
