I've got a 3.5 inch internal harddisk . I will use this harddisk mainly for storage. It's 2T . Do I really need to partition ? Does partition have any positive or negative effects to the harddisk itself?

Solution
You have a partition no matter what. It is just the entire disk is a single partition. A partition is a necessary ingredient in organization of the disk. Now, if your question is do you need MORE THAN ONE partition, that is a slightly different question. IF you have only a single disk, then splitting it to say 200GB for a C: drive and the rest for a D: drive makes it easy to clone the C: drive to an SSD when you have the $$$$.

Making your windows partition smaller may a have speed benefit. I remember reading something about clusters or sectors being smaller if you use a smaller partition, thus making certain read/writes more efficient.

If you don't have any need for a system portion, then a single partition is perfectly fine. Unless you have a reason, most people don't, to limit the space usage for an application or to isolate something there is no reason to not use a single partition.
