Okay now some more details.

1. When you power up the computer, does anything show on screen at all?
2. Does this GPU have additional power to plug in- for example 6pin?
3. Can you remove the GPU and plug it back in? Make sure it is in tight and you shouldn't be able to see any gold pins.

Okay the fact that you can see anything once you hit the power button is good. It means the hardware is functioning at this level.

So what do you mean it doesn't boot up? Do you just mean you can't get into Windows?

If so, boot to Safe Mode. See if you can get that far.

Usually that is because of a bad boot sector, if I remember correctly.
Flashing _ is looking for the HDD and the partition.

You can get into your BIOS on the motherboard, most likely the F2/setup key.

Try resetting all the options.
Make sure your HDD is the first boot option after resetting.

If that doesn't work, put the Windows OS disk into the disk drive and see if it can load the setup. Don't actually do the setup, I just want to see if you can do it.

He can't get into bios.

Put your os disk in the DVD drive. Can you get to the installer?

If so the issue is with the HDD.

Are you sure you didn't accidentally disconnect the HDD connectors on the back of the HDD or the motherboard?

I came across this while researching your problem.

As per troubleshooting steps, please try this:

Clear the CMOS by removing all components from the board (power supply, battery, hdds) for about 20-30 minutes and then reconnect them again.

I tried removing the CMOS battery and put it back again after 10 minutes then after startup there are 3 short beeps 1 and 2 beeps have at time delay of 25 sec and third and fourth beeps have a delay of 33 sec and then the screen goes blinking. Also when I remove the graphics card the computer boots up normally and every thing is fine also I can access bios.

Sorry if I wasn't clear but, remove the graphics card and connect your monitor via the on-board graphics solution. Boot up and access the bios and look around for graphics settings. Usually, you can specify which device you want the computer to use by default. Set this to external or whatever is there and try that.
