There is no significant difference as far as I know. You'll lose some bandwidth if you go with the single 8gb ram stick. The dual 4gb ram stick is a bit faster but is really unnoticeable. But, if you have two 4gb ram sticks, you'll only be able to expand to 16gb. While if you have one 8gb ram stick, you can expand up to 32 gigabytes of ram.

The computer can work with either configuration. There is a slight performance advantage with the 2 x 4 GB over the single 8 GB stick due to the dual channel memory effect. It is about 5-10% faster for the same memory with the dual sticks over the single stick.
