Can I use my passport to backup a thumb drive rather than my hard drive? How?

You could try using the WD SmartWare backup software that comes with the WD My Passport to target your flash drive for backup.

However, you should keep in mind that it would need to be re-formatted in NTFS in order for SmartWare to pick it up as a target.
If you decide to do this, make sure you already have a backup of the data that's on the thumb drive, since re-formatting would wipe it.

Or a manual copy/paste should work as well!

It's no different to copying files from one hard drive to another.
Plug in your pen drive and your Passport.
Open a separate window for each drive.
Place windows side by side.
Drag-and-drop files from one window to the other.
