Can I store AppData on a separate drive from the OS

So my new SSD arrived today and so I'm clearing out space for a cloning to it. AppData in my user folder is taking up about 26GB. Can I copy that over to another drive and store it there or would there be issues?

I do understand it would have to stay with the Users folder so I guess the question is can I store users on a separate drive.

Well the SSD size is more than enough right now really. I have a 250gb(232 in reality) and I only have about 91gb left on my main drive. I know that as SSDs fill up they become slower so I was wanting to get rid of as much as I could.

Okay, understood. After searching through my programs I found some other stuff to get rid of and got it down to 78.8gb. Thanks for your help.
One last question if you're still here, should I defrag my HDD before cloning? I assume yes but no harm in asking.
