Gigabyte GA-Z97X Gaming 3 in Corsair Carbide Series 300R

The Gigabyte GA-Z97X Gaming 3 has 7 mounting holes:

o o
The red one doesn't line up with 3rd row of ATX format so it's useless. Moreover in the place of the blue one my case has positioning bolt so I won't be able to screw it there.

Will it be rigid enough considering I'm going to install an aftermarket CPU cooler?

From what i can see over the 300R, there shouldn't be any problem with blue hole at all, i don't get whats the
issue.

With the red one there might, but with some mobos or cases you get plastic mounting posts like here on the screen (those creemy like), in which case you don't need any hole at all.

Btw CPU cooler will be fine without the red one, I'm more concerned about pushing RAM sticks.
Without the blue one also, mobo could also bend during the montage of CPU, depends exactly how much pressure it will be required.... its not likely you gonna break it if you are gentle enough.
