Password protect/lock folder on portable hard drive?

Is there a way to lock/password protect only one folder on a portable hard drive and still be able to transfer files between a Mac and Windows computer?

You can create encrypted folders using TrueCrypt and there is a MAC version. While there is a warning about it not being secure, that is not too worrisome for most users, you are not likely to be saving the data of 100,000 credit card numbers and passwords that someone would want to hack into.
