I have tried installing the latest realtek audio driver from the asus websites.

When I plug in my headphones/speaker cable into the green audio on rear, it detects it and says headphones have been plugged in and automatically makes the speakers default in playback settings. But no audio comes through.

any suggestions?

I also get no audio from the front panel.

Solution
Update your motherboard BIOS (search your model on the manufacturers' website).

Download Driver Booster 3 and install/reinstall any missing/outdated drivers.

Restart your computer then again try the steps I mentioned in my first reply.

Right click your sound icon in the bottom right hand corner, select Playback Devices, disable your speakers, now in a blank area right click and select "Show disabled devices" and enable your headphones and set it to your default device.
