May I know is this CPU able to support for playing Dota 2 and do some working like Microsoft?

I think Q6600 will run DOTA2 just fine as long as you have enough RAM and a good GPU.
I would upgrade the RAM to 8GB and use GTX750Ti for GPU.
Do not go higher than GTX750Ti, your Q6600 is a bottleneck to most new games for GPUs higher than GTX750Ti.

For office daily works like microsoft office, Q6600 is more than enough.
