Actually it should they are both listed as 73W TDP. You will need thermal compound to re-apply the heatsink to the 'new' processor.

$50 or $65 is quite a bit of money towards a 5 year old CPU. 1st Gen i5 600 are dual cores with hyperthreading. Basically the i3 of today. And honestly not all that different from what you have now. A slightly better upgrade would be this true quad core:

That CPU would require a new CPU cooler as it is listed at 95W, and is risky to install in that motherboard.

i7-870 are going for closer to $100, and that is money that could be spent on actually...

Not going to be easy to find a 1st gen LGA1156 processor. Best possible would be an i7-880 or the like, but since that is an OEM motherboard there is no absolute guarantee it would work.

Better off just buying the GPU and PSU now. Or buying a standard case so you can get a standard power supply rather then the small form factor one so that you can upgrade the motherboard and CPU at a later date.

Would the CPU cooler that came stock with my machine work for this CPU?

I would leave the CPU alone and save your money. GTX750 is a decent entry level card that will get you medium settings in most FHD games, a little below the minimum 750Ti or 950 that is current entry level choice.

If instead you were to invest in a standard power supply and a new case, you could then use full size dual slot cards. There are a few GTX750 and 750Ti that are clocked much higher then the low-profile ones for only $10-20 more.
