For my computer class, we had to download VirtualBox and install Windows 8.1 Pro. I have the .iso and the product key. However, after the class was finished, I forgot to uninstall/remove the product key before deleting Windows from VirtualBox. Is it still possible for me to use that product key for my own Windows 8.1 computer and upgrade it to Pro?

If the key is retail and you were told it's yours to keep, sure. If the key was OEM, you wouldn't have been able to anyway. If the key was a volume license key, you might get kicked out of school or forced to compensate the school for theft of a key.
