Does it matter what Sata port I use?

From what I'm seeing in the mobo manual, all ports are SATA 3 ports except for the 2 SATA express ports. So will it matter which ones I plug my sad, main storage drive into?

From what I understand it won't matter but want to ask the experts.

What's a sata express port?

Edit: When I do my install, should only the DVD drive and SSD be plugged in or can I have my 3 storage drives up and running too?

No, it won't matter.

A SATA-Express port it two SATA-3 ports, which can either be used separately to connect two normal SATA devices, or bound together with a single SATA-Express cable to power a SATA-Express device (essentially it's a PCI-E x2 connection).
