Old SSD & HDD to one new SSD

I have my upgraded Windows 10 & some other things on a Samsung 60gb 830 SSD ( C drive ). I also have an HDD (I drive ) that contains most of my other programs & files.
I want to replace both with one 500gb Samsung 850 SSD.
Can the two old drives be combined on the new SSD with drive letter C?
If not, can I copy each of the old drives on to the new SSD using the Samsung migration software?
Would the new SSD run any slower having to use two drive letters?
I would prefer the first option, in the hope that it may remove duplications of data, that may be on both drives, basically clean them up.

You could try cloning the the SSD first. After that make sure that the clone was successful and that you are able to boot to Windows with your new drive. If this has been achieved you can extend a bit the partition e.g. to 100-120GB so that you have more space for your C: drive. Leave the rest of it as unallocated space. Now clone the HDD to the new SSD. Note that I'm not sure which software you should use in order not to format your whole destination drive with the 2nd cloning process so you'd have to look into that. If you are successful with both partitions, make sure that the 2nd partition matches the letter of your old drive so that all paths are correct for the programs and files.

Just one thing to add to Boogieman's response since you asked about the possibility...

Assuming you've been able to manage a successful disk-cloning operation so that your 500 GB SSD now contains the two partitions from your other SSD & your HDD, it is possible to merge the two partitions into a single partition (C:\), however you would need to employ a commercial third-party partition management program to accomplish this. While most of those types of programs have "freebies" available, I am not aware that any of them possess this type of merging capability. At least I haven't found any.

Again, assuming you've managed a successful disk-cloning operation the likelihood is you'll have to live with two partitions. And do follow B's advice to assign the same drive letter to the partition you cloned from your secondary HDD.
