Setting up a RAID do I have to have my OS SSD connected?

I'm trying to set up a RAID configuration with 4 3T HDD's. I currently use a Samsung SSD as my boot drive and storage for programs. The other 4 drives I want to use for storage and redundancy.
What I want to know is can i just disconnect my Samsung SSD and configure the RAID and after plug it back in?
In the BIOS right now it is showing my samsung as one of the disks for the RAID config so I stopped here for fear of erasing all my data.

It doesn't give me the option of selecting the disks it just ask what type of RAID array and then shows me the disks available. I'm using an ASUS z170 Pro Gaming board ASUS Ez Tuning wizard.

O,K., don't use Asus,s software. Go into the BIOS RAID splash screen or Disc management in the OS.
The RAID splash screen is f5 I think. Ck your manual. It will come up before the BIOS screen. Hit f5 instead of enter or delete for BIOS entry. Activate RAID on the BIOS screen first and then do over for the RAID screen.

Yes just using the easy tuning in ASUS and unplugging my system drive (SSD). I wanted to do a RAID 10 but it seems that would be the equivalent to the "two way mirror"(windows 10) which I will most likely do. The three way mirroring seems the safest as far as back up but that requires 5 drives.
