Can I use one of my P8Z77-V deluxe PCIe 3.0/2.0 x16 slots for a Addonics M2 PCIe SSD - PCIe 3.0 4-Lane Adapter with Samsung 950 Pro M.2 SSD? If so will it utilize the fast r/w speeds of the Samsung 950 m.2 and not effect esata throughput?

NVM Express (NVMe) or Non-Volatile Memory Host Controller Interface support was provided to the Asus Series 9 chipset motherboards. Asus has no plans to provide NVMe support for legacy Series 7 chipset motherboards such as the Z77 chipset P8Z77-V.
