Will a GTX 1070 work perfectly with a ASUS H81M-C motherboard???

Yes. The motherboard just has a standard PCIe slot that's fine, and the CPU and RAM are appropriately powerful for that kind of graphics card.

The one thing you haven't checked is the power supply, which needs to have sufficient wattage, 12V amperage, and power connectors to support the card, and also be decent quality. But the 1070 isn't that power-hungry, so there's a good chance you're fine.
