I've just finished assembling my build, i haven't updated the bios yet as i couldn't find option. installed windows 10 pro on ssd. Everything looked like it was running smooth and very quick aswell actually. Now i'm going to plug back in the hard drive....but what steps or advice would you now give? E.g make a partition or just inititialize and quick format. What else to optimise such as any particular settings? Once this has been completed i shall upload pictures shortly of my pc gaming rig and also pictures of temperature and load benchmarks.

Your old HDD will have some partitions which you don't need any more and that space could be used again. After you install it, you can run "diskmgmt.msc" , delete all the partitions and make new one or more formatted to NTFS and assign it/them a letter each.
Another possibility with more options would be to use http://www.pcdisk.com/download.html for same purpose.
