If I understand correctly, is it:

24-Pin PSU plug
4-Pin CPU plug
CPU
Stock cooler/aftermarket cooler
RAM

Is there anything more (or less) required?

I don't care about video output for the post test since I just want to test if the CPU fan and case fans spin which is progress for me since my PC would only spin once then stop.

Thank you!

Yes that would be a basic bare minimum test Gilbert.

The system not powering up right if all the correct required power from the Psu is connected to the motherboard though.
Is an indication that the power supply of the system if old is failing.

If the current Psu in the system is over three years old or more.

If the system was working, and was on at the time you last used it but you had a power cut, or an electrical storm that caused power to be lost.

Then the reason as to why the power supply is not powering the system up properly when the power switch, or button is pressed on the system.

May be because one of the protection circuits of the Psu has been triggered, and needs to be reset using a set procedure.

If that fails to...

If that fails to resolve the power up problem of the system, then you should try the system with another power supply unit connected to it.

It's a first build that hasn't turned on at all yet. Replaced mobo and still didn't work. About to try out my new PSU replacement. I will keep all of that in mind for the future though, so your advice is appreciated!
