Amount of ram cards being used makes a difference?

so im building a gaming pc for the first time and i am wondering if it makes a differencee if i put 1 8gb ram or 2x4gb ram

Not really no, depending on the number of ram slots on the motherboard though I would recommend 8gb because then it gives you an option to upgrade to 2x8gb without buying two new sticks to replace the 2x4gb sticks. You just need to add another 8gb one.
