So I know this might seem like a dumb question too some but I honestly have no idea. So my question is will the GTX 1080 be able to run Borderlands 2 @1080 144Hz and not go below that on max settings with Physx On or Off either one I don't really notice it to be honest and I understand that if you have a 60hz monitor this is no question for you this card will destroy 1080p but for 144Hz I don't know thank you anyone willing to respond ;)

I think it should be able to with no problems, you will have turn down some setting, maybe Physx as you said, with that, It should work properly at 144hz. On action full moments, It might drop a bit tho.

With physx set to high, a gtx1080 will occasionally go below 20fps in multiplayer even with a 640-480 resolution.

Physx in most games is like a tech demo poorly implemented and optimized. The only thing optimized about physx is their ads.
 
Setting Physx to high is silly, takes a huge performance hit and you don't even notice it.
For example in the Metro Redux series which I recently got around to playing, there's an option called advanced Physx, and after turning it on, my framerate went from 120 maxed at 1440p to 30-40.

On my GTX1070, setting physx to medium results in almost no improvement in performance but it removes some physx effects in BL2 altogether (like liquid spilling from ceilings). The only way to play the game without droping as low as 15fps is to set physx to low.
