Can I use another GPU if I have an Integrated Chipset?

I have an Intel G43/G45 Chipset which I want to replace so I can run games like Skyrim. Can I buy a different GPU and use that one instead? If so, how would I go about doing this? The GPU I'm looking at ordering it this one: https://www.amazon.com/gp/product/B004MSQKLO/ref=ox_sc_act_title_1?ie=UTF8&psc=1&smid=A2PY6XR1EZ5L0D

(Don't have much money, so buying on a budget.)

My Motherboard is also this one: https://www.asus.com/Motherboards/P5QLVM_EPU/ if it matters.

You don't need to do anything to use a dedicated graphics card. Just put it in the x16 slot and plug the monitor into it (instead of the iGPU). Is $45 w/shp the limit for a gfx card?

Yeah about $45. Having money troubles now, but had a small extra bit. I can always wait until Christmas when I get more. I prefer Amazon over EBay, less chance of getting scammed, even if you pay a little extra.

I'm afraid you're not going to be happy with that older HD-HD6350 playing Skyrim. That is about the minimum requirement for that game. If you can squeeze out a few more bucks, this will improve the graphics threefold.
